- W4312552694 abstract "A sensitive liquid chromatography method was developed for the determination of β-carotene in high β-carotene modified cassava flour (MOCAF). β-carotene was separated isocratically on a reversed-phase high performance liquid chromatography (HPLC) system, employing a C18 column stationary phase and a methanol-acetonitrile elution system with the ratio of 3:7 as mobile phase. The flow rate was maintained at 1.2 mL/min and the response was detected using a diode array detector (DAD). The validity of this method was confirmed by linearity, precision, detection limit, and quantitation limit. Results of the present study show that the method has excellent linearity in the range of 0.07 to 11.52 µg/g, with the coefficient correlation of 0.9997 and detection limit at 0.02 µg/g. The intermediate precision, expressed in RSD value of β-carotene peak area, was found to be 1.01%. The validated HPLC method was also successfully utilized for the estimation of β-carotene content in high-carotene MOCAF derived from cassava rich in β-carotene cultivars Mentega 2 and Carvita. This indicates that the method is suitable and reliable for routine procedures. Furthermore, this method can be applied to various types of flour and processed products. The method is particularly beneficial for the measurement of β-carotene, which was regarded as too low to be detected in MOCAF samples." @default.
