By switching fluorophores between two distinctive states, super-resolution fluorescence microscopy overcomes the diffraction limit and revolutionizes light microscopy with its capacity of visualizing subcellular structures and organelles at the macromolecular level. In this contribution, we discuss stimulated emission depletion (STED) and ground-state depletion (GSD), two mechanisms that are widely used to reach super-resolution.
