- W4313309637 abstract "In this study, a systematic approach was established in order to define and optimize the required amino acids in a defined minimal medium for the synthesis of human interleukin 3 (rHuIL-3) in Streptomyces lividans . Initially, the number of probable required amino acids was narrowed down from 20 to 8 using starvation trials. Starvation trials were followed by designing and performing screening mixture experiments on the identified eight amino acids. In addition, the eight probable amino acids were ranked based on both growth and product formation using distance-based multivariate analysis. The application of previous steps facilitated narrowing the search from eight to only four amino acids (aspartic acid (Asp), leucine (Leu), methionine (Met), and phenylalanine (Phe)). A mixture design experiment called simplex lattice design was conducted to identify the composition of optimum minimal medium. The results revealed that the acids composition of Asp 53%, Met 5%, and Phe 42% would deliver the optimum minimal medium.
