- W4313426686 abstract "Cellular imaging of thick samples usually requires either physical sectioning or laser scanning microscopy, which are time-consuming and involved. We developed a compact fibre optic-illuminated imaging platform based on microscopy with ultraviolet (UV) surface excitation (MUSE) for cellular visualisation of thick sample surfaces. Optical fibres delivered illumination at steep oblique angles, optimising optical sectioning by minimising UV penetration depth, and enabled short-working-distance objective lenses. Multi-scale image mapping spanned single cells to murine organ slices of 8x12 mm and 1.3 million cells per surface. Omni-directional control of optical fibre illumination produced 3D microtopography, a new concept relevant to microstructure and heterogeneity studies. We demonstrated histological imaging validated against standard H&E, multiplexed fluorescence for protein expression, and quantitative mapping of cell and organ-scale viability including an analysis of antibiotic insult on ex vivo murine kidneys (n = 12). This technology addresses long-standing gaps in laboratories for visualising thick samples for near-real-time cellular insights." @default.
