- W4318773714 abstract "Transcriptional factor binding and histone modifications play key roles in biological processes. With the advance of chromatin immunoprecipitation followed by sequencing (ChIP-seq), we can profile the chromatin regulatory landscape of cells and tissues and their associated transcription regulatory network. ChIP-seq is the prototype and the most widely used techniques for mapping genomic distribution of transcription factors, histone modifications, and other chromatin modulators. After the isolation and shearing of chromatin, antibodies to the antigen of interest are used to pull down associated DNA, which is then mapped to the genome to determine the DNA sequence the protein binds to, as well as its genome-wide distribution. ChIP-seq can be performed with cultured cells or tissues, in a spatial and temporal manner. However, the semi-quantitative nature, as well as the multiple sources of biological and technical variability inherent to traditional ChIP-seq technology prevent cross sample comparisons. Moreover, conventional ChIP-seq protocol is not suitable for clinical settings because of the demand of large amount of starting materials and low resolution. Herein, we summarize the advantage and limitations of traditional ChIP-seq method and provide an overview of evolution of ChIP-seq protocols in chronic order Key, with improvement on reducing sample material and enhancing resolution.
