- W4319787054 abstract "My name is Bianca B. Batista. I was born in Ribeirão Preto, São Paulo, Brazil. I am currently in the last year of my PhD in the Cellular and Molecular Biology program in the Ribeirão Preto School of Medicine at the University of São Paulo. Employing an unbiased approach, our goal is to identify transcription factors, besides Fur (ferric uptake regulator), involved in iron homeostasis in the environmental opportunistic pathogen Chromobacterium violaceum. I work under the supervision of Prof. José Freire da Silva Neto, an Associate Professor at the University of São Paulo. We identified six possible regulatory systems, besides Fur, involved in iron homeostasis in C. violaceum using transposon mutagenesis and PSA-CAS plates. Notably, deletion of the CV_1057 transcriptional regulator led to an increase in siderophore halos, as well as a decrease in biofilm, violacein, protease, and hemolysin production. Further characterization showed that CV_1057 regulates the expression of vioS, a divergently located gene within the CV_1057 genetic cluster, as seen using RNAseq. Interestingly, the CV_1057 regulon overlapped the CviR regulon, suggesting that CV_1057 might act upstream CviR. Indeed, insertion mutations in cviR and null mutation of cviI (the gene that encodes the production of the autoinducer molecule) and cviR led to an increase in siderophore halos. Genes related to iron homeostasis and those encoding the CPI1 T3SS were regulated by CviR but not CviI, suggesting that CviR can also act without its canonical CviI autoinducer. Our data revealed a complex regulatory cascade involving QS that controls iron homeostasis via siderophore-mediated iron uptake in C. violaceum. My next step after defending my PhD is to apply for a postdoc position. I am certain this is a great opportunity for me to learn new techniques, gather teaching experience, and do networking.
