- W4324141874 abstract "Purpose: Joint damage caused by trauma can proceed to post-traumatic osteoarthritis (PTOA) due to the limited tissue repair capability. Currently, there is no cure available to prevent progressive joint degeneration, therefore there is an immediate need to develop treatments to prevent PTOA. Systemic pharmacological treatments can cause serious side effects and there is a need in the field to develop joint local therapies. Synovial macrophages play a role in OA onset, progression, and pain. In previous studies, we monitored macrophage infiltration in two different PTOA mouse models, anterior cruciate ligament transection (ACLT) and the destabilization of the medial meniscus (DMM), using a macrophage targeted nanoemulsion that contains a near-infrared fluorescent marker (provided by Dr. Janjic’s group). We established that macrophage infiltration increases in injured knees and is maintained during PTOA progression in both models, with ACLT exhibiting higher level of cell infiltration. Previous work by Low’s group demonstrated that activated macrophages express a folate-receptor-β (FR-β) that is not expressed by resident non-activated macrophages; therefore FR-β can mediate the internalization of folate-linked molecules, such as imaging agents or therapeutic drugs. Kraus et al. used an imaging technique through FR-β binding to monitor activated macrophages in knees of OA patients and reported that the quantity of activated macrophages correlated with radiographic OA severity and symptoms. Other reports suggest that targeting FR-β might represent the key for potential therapies in arthritis. We recently established a collaboration with Dr. Low and obtained a FR-β-targeted near-infrared contrast agent (OTL38) to analyze the in-vivo distribution of activated macrophages following rat ACLT with the purpose of establishing whether activated macrophages can be used as drug carrier for potential PTOA therapies." @default.
