However, conventional therapies for melanoma management face several drawbacks, like poor permeation across the skin, poor accumulation in the tumor, and chemoresistance. We have developed Pioglitazone-loaded liposomes (Pio-Lipo) embedded within the hyaluronic acid gel owing to acidic pH-responsive depot formation. The liposomal formulation was optimized by employing quality by design. The formulated liposomes had a particle size of 134.87 nm, PDI of 0.157and a high entrapment efficiency of 92.17%. Intending to satisfy the unmet clinical needs and emphasize the concept of drug repurposing, we have employed Pioglitazone, an approved anti-diabetic drug, to manage melanoma. In vitro release kinetics demonstrated sustained release of the drug from the gel matrix till 24 h, where the release of free drug and formulation followed zero-order and diffusion-based release mechanisms. Cellular internalization studies confirmed the successful internalization of Pio-Lipo in SK-MEL-28 cells. Nuclear staining study revealed nuclear fragmentation and blebbing of the nucleus, indicating cell apoptosis. To assess cell viability post-treatment, an MTT assay was performed, where Pio-Lipo displayed greater cytotoxicity towards SK-MEL-28 cells (P < 0.05) compared to free Pioglitazone. Thus, Pioglitazone-based liposomal gel could be an effective alternative for melanoma management." @default.
