The Atopic Dermatitis (AD) is an eczematous skin disorder affecting 15-25% of children and 7-10% of the adult’s population. In atopic dermatitis, the main characteristic is a dysfunction of the skin barrier with a strong decrease of differentiation markers including a dysregulation of lipids organization at the lesional skin site. The development of specialized human skin reconstructed models is a very adapted tool allowing to better understand Atopic Dermatitis. We developed epidermal reconstructed skin models in which we induced characteristics of AD. Indeed, the 3D models are developed using a well-defined culture medium and with the application of a combination of cytokines and glucocerebrosidase (an enzyme of the ceramide pathway) gene silencing on keratinocytes, in order to induce barrier dysfunction and immune characteristics of AD. The study of the modulation of Atopic Dermatitis related markers was deepened by immunostaining on the 3D models. The AD characteristics were identified in the reconstructed epidermis model. Indeed, the barrier function was affected in these reconstructed models. Actually, the thickness of the tissues was impacted by cytokines application and glucocerebrosidase silencing. Several proteins of the epidermal differentiation complex are downregulated such as filaggrin, a main gene dysregulated in the AD. The balance of lipids, the cohesion and the thickness of reconstructed epidermis were also modulated. Our reconstructed epidermis is an Atopic-dermatitis-like model with a well-defined barrier dysregulation induced by specific regulators of the disease. This type of reconstructed skin model is a fine-tuned tool to evaluate atopic skin characteristics such as the AD skin disorder. This kind of bioengineered skin models will be used to screen and identify compounds and ingredients with beneficial effects on the disturbed skin barrier and inflammatory signals delivered in Atopic Dermatitis." @default.
