- W4366181617 abstract "In postoperative wound care after surgical procedures or ablative laser treatments, recent clinical studies suggest the early use of non-ablative fractional laser treatments to improve wound healing. This technique is called laser-assisted scar healing (LASH). However, the underlying molecular mechanisms are still poorly understood. The aim of this study was to investigate the biological effects of LASH and to combine it with already established topical treatments that promote wound healing. We first irradiated 3D skin models with a fractional ablative Er:YAG laser to set standardized lesions. Next, we treated these models with either a non-ablative fractional 1540-nm Er:glass laser or 1550-nm diode laser (LASH approaches). In an additional approach, we investigated the combination of LASH with direct topical post-treatment using a dexpanthenol-containing ointment (DCO). Histologic analyses showed that models treated with LASH exhibited an accelerated wound closure after 16 hours, with lesions still visible. In contrast, an additional topical post-treatment with DCO led to complete wound closure. Microarray analysis revealed that LASH stimulated an upregulation of epidermal differentiation markers (e.g. FLG, LOR) and showed mild anti-inflammatory effects. Post-treatment with DCO enhanced the wound healing effects of LASH, especially the anti-inflammatory effect. In conclusion, ourin vitrostudy showed that LASH treatment with a fractional non-ablative 1540nm Er:glass or 1550nm diode laser improved wound closure in the early phase of wound healing by exhibiting similar biological effects. Post-treatment with DCO promoted the proliferative and anti-inflammatory effects of LASH and is therefore a useful addition. These data contribute to a better understanding of the biological effects of the LASH technique and help to optimize its application." @default.
