- W4366181661 abstract "Skin resident memory T cells (TRM) are a subset of memory T cells which reside in skin for a long term. They require exogenous free fatty acid for their survival, and fatty-acid binding protein 4 (FABP4) and 5 (FABP5) are considered to take part in lipid uptake. However, the detailed relation of FABP4 and 5 with TRM function is still unknown. Here, we analyzed the role of FABP4 and 5 in the development and functional activities of skin TRM in a murine contact hypersensitivity model. Wild type (WT), FABP4 knockout (FABP4KO), and FABP5 knockout (FABP5KO) mice were sensitized on the abdomen with 0.5% DNFB at day -7 and challenged on the right ear with 0.15% DNFB at day 0. By day 28, CD69+CD103+CD8 TRM develop in the right ear. When both ears were re-challenged with 0.15% DNFB at day 28, the swelling of the right ear at day 29 compared to left was the weakest in FABP5KO, followed by FABP4KO and WT (0.146 ± 0.007 mm in WT, 0.094 ± 0.009 mm in FABP4KO, 0.052 ± 0.007 mm in FABP5KO, WT vs FABP4KO: p < 0.001, WT vs FABP5KO: p < 0.001, FABP4KO vs FABP5KO: p < 0.05). The number of CD69+CD103+CD8 TRM in the right ear at day 28 tended to be lower in FABP4KO and FABP5KO than in WT, however, the difference did not reach significance (484 ± 65 cells/ear in WT, 364 ± 41 cells/ear in FABP4KO, 340 ± 52 cells/ear in FABP5KO, WT vs FABP4KO: p = 0.3304, WT vs FABP5KO: p = 0.1240, FABP4KO vs FABP5KO: p = 0.9980). IFNγ production from this fraction was comparable in FABP4KO and WT, while that of FABP5KO was decreased compared to FABP4KO (104 ± 18 cells in WT, 147 ± 19 cells in FABP4KO, 58 ± 11 cells in FABP5KO, WT vs FABP4KO: p =0.2107, WT vs FABP5KO: p = 0.2067, FABP4KO vs FABP5KO: p < 0.05). Our results imply that FABP5, rather than FABP4, contributes to both the development and functional activities of skin TRM." @default.
