- W4366182196 abstract "Previously, we discovered that noncoding double-stranded RNA (dsRNA) is secreted from damaged skin and dsRNA-induced TLR3 activation stimulate intrinsic retinoic acid synthesis to promote regeneration. Using genome-wide microarray analysis to investigate the novel mechanisms, we identified that the genes involved in exosome biosynthesis were upregulated in human keratinocytes treated with Poly I:C, a synthetic dsRNA. Basically, exosomes are a type of extracellular vesicles (EVs) secreted from cells, and contain biomacromolecules such as polypeptides, lipids, miRNA, mRNA and DNA. Exosomes can be fused with other cells to deliver the materials. Given that, we hypothesized that dsRNA released by damaged skin will be internalized through exosomes and successfully delivered to endosomal TLR3 to promote tissue regeneration. In this study, using TLR3-siRNA, we confirmed that TLR3 activation is essential for the expression of exosome markers, which leads to the regulation of exosome biosynthesis. In addition, we found that the expression of hair follicle stem cell markers (KRT15 and KRT19) was significantly increased in keratinocytes treated with TLR3 activation-induced exosomes. To investigate bioactive molecules in exosomes, we used metabolomics and RNA sequencing. Metabolomics results show the increased contents of threose and alanine in TLR3-activated keratincoytes. Also, RNA sequencing analysis results showed that miR-193b-5p was abundant in exosomes induced by TLR3 activation. Finally, we expect that novel bioactive molecules will be discovered and developed the functional materials to promote tissue regeneration through further investigations." @default.
