- W4366185375 abstract "Solar lentigo (SL) is a common hyperpigmented lesion occurring on sun-exposed skin, and is a sign of photodamage and aging. WNT/β-catenin signaling promotes melanogenesis via activating MITF, the master regulator of melanocytes. Secreted frizzled related proteins (sFRP) are antagonists of WNT/β-catenin signaling that inhibit the binding of WNT ligands to their receptors. However, recent studies have reported that sFRP2 activates melanocytes, suggesting opposing effects of sFRP proteins. In this study, we focused on sFRP1, whose function in melanocytes is unclear, and examined its regulation of melanocytes and possibly SL formation. Paired skin biopsies were obtained from SL lesion and peri-lesional (PL) skin of female volunteers aged 50-68 years. Dermal RNA was extracted and the expression levels of sFRP1 were measured with real-time qPCR. The effects of recombinant protein of sFRP1 (rsFRP1) were determined on the morphology, cell number, viability, and melanogenesis of cultured normal human melanocytes. The expression levels of sFRP1 in the several types of cultured skin cells were determined by real-time qPCR, Western blotting, and ELISA. The mRNA expression level of sFRP1 was decreased in SL areas compared to PL areas. Treatment of cultured human melanocytes with rsFRP1 decreased their dendricity, proliferation, and tyrosinase activity. Expression of sFRP1 was highest in keratinocytes, and evident in fibroblasts, and dermal papilla cells. These results suggest that sFRP1 is produced by different skin cells and negatively regulates melanocytes, and that its reduced dermal expression in SL is involved in the pathogenesis of SL. Enhancing sFRP1 expression in skin may be a potential therapeutic approach for hyperpigmentation." @default.
