- W4366185410 abstract "Hyaluronic acid/Hyaluronan (HA) is an important component of the extracellular matrix of dermis. HYBID (HA-Binding protein Involved in HA Depolymerization), also known as KIAA1199/CEMIP, plays a key role in the degradation of large HA (>1000 kDa) into intermediate size HA fragments with molecular weights of 10 kDa to 100 kDa. Recently, we have demonstrated that Artemisia capillaris flower extract suppresses the histamine-induced expression of HYBID in neonatal normal human dermal fibroblasts (NHDF). In this study, as HYBID is responsible for the degradation of HA, we have evaluated the effect of the same plant extract on the protection of HA from degradation under the same experimental conditions. NHDF were maintained in Dulbecco's Modified Eagle Medium (DMEM) containing 10% fetal bovine serum. After the cells were treated with different concentrations of Artemisia capillaris flower extract for around 3 hrs, they were exposed with a HYBID inducer, histamine, for an additional 3 hrs followed by the addition of fluorescein isothiocyanate (FITC)-labeled large HA (1,200-1,600 kDa). After 24 and 48 hrs of incubation, the cultured medium was collected for the measurement of the molecular size distribution of FITC-labeled HA by HPLC. It was found that not only the large HA was decreased, but also the intermediate size HA was increased in the medium of cultured NHDF by the effect of histamine-induced HYBID. In contrast, the use of Artemisia capillaris flower extract time- and dose-dependently reversed the action, which leads to the protection of large HA from degradation. As the loss of large HA has been reported to be directly correlated with wrinkle and sagging of skin, Artemisia capillaris flower extract could provide an important and non-invasive solution to protect our skin against aging through protection of HA from degradation by HYBID." @default.
