- W4366185416 abstract "The developmental origins of health and a disease (DOHaD) hypothesis, which was formulated during the Dutch famine at World War II, provided the first, direct evidence in humans of the lasting consequences of prenatal condition. This concept postulates that the environment during early development in utero, and early post-natal periods may be associated with the development of non-communicable diseases, such as diabetes and cancer. The present study is the first to develop an animal model of atopic dermatitis based on the DOHaD approach. In the present study, we transferred NC/Tnd mice, which are registered internationally as a spontaneous murine model for atopic dermatitis, from the specific pathogen-free (SPF) environment to the conventional environment and changed the breeding period. We investigated the incidence of dermatitis and found that in the course of pregnancy, childbirth, and weaning, the longer the time spent in the conventional environment of the mother mouse, the less likely were the pups to develop dermatitis. The microbiota on the skin and intestinal tract regulates the function of immune cells via the metabolites produced to maintain homeostasis. Disruption of this microbiota contribute to the development of various diseases, such as diabetes, cancer, autism, and arteriosclerosis. The microbiota of skin without dermatitis was highly diverse, and the metagenomic analysis using 16s ribosomal RNA revealed that the microbiota in mice with and without dermatitis differed markedly. Some bacterias having a positive correlation with IL-4, IL-5 and IL-10 and others having negative correlation with those cytokines were able to be identified. These results newly identified a link between microbiomes and T cell function. The present study is the first to develop an animal model of dermatitis based on the DOHaD approach that is capable of prenatally suppressing atopic dermatitis." @default.
