- W4366185738 abstract "Alopecia areata (AA) is a common hair loss condition characterized by the acute onset of scarring hair loss. Recently, JAK inhibitors have been used to treat patients with severe AA who do not respond to conventional treatments such as glucocorticosteroids, cyclosporine, and diphenylcyclopropenone. Our previous studies showed an alternative effect of JAK inhibitors on hair follicle cells, decreasing mitochondrial damage and inflammasome activation, thereby improving AA's inflammatory status. However, no reports have investigated the role of JAK inhibitors in direct hair growth promotion in hair follicle cells. Therefore, this study investigated the effect of JAK inhibitors on β-catenin activation in human primary outer root sheath (ORS) cells. IFNγ-treatment decreased the β-catenin and increased Dickkopf-related protein 1 (DKK1) expression in ORS cells in a dose-dependent manner, and β-catenin was decreased in both cytosol and nuclear fraction. Next, we used each selective JAK inhibitor, itacitinib, CEP-33779, and ritlecitinib, to examine which JAK inhibitor regulates β-catenin. Itacitinib and CEP-33779 increased β-catenin in the cytosol and nucleus of ORS cells which was inhibited by IFNγ treatment and ritlecitinib slightly reversed β-catenin compared to other JAK inhibitors. Moreover, itacitinib and CEP-33779 increased transcriptional activation of β-catenin in ORS cells in a dose-dependent manner. Therefore, this study shows that JAK1 and JAK2 inhibitors increase β-catenin activity and decrease DKK1 levels, thereby providing an additional therapeutic mechanism of JAK inhibitors other than inhibiting the JAK-STAT pathway and gives more information to dermatologists to use JAK inhibitors in patients with severe AA." @default.
