- W4366186280 abstract "Staphylococcus aureus (S. aureus) is often found in the skin of patients with atopic dermatitis (AD) but also in patients with irritant contact dermatitis (ICD) such as hand eczema. Hand eczema is known to be a common complication of AD, and to be more severe with the colonization of S. aureus. These results suggests that skin-colonized S. aureus possibly aggravate the cascades of ICD. However, it is not conclusive that the presence of S. aureus is a cause or a consequence of the ICD exacerbation. The purpose of this study is to clarify the causal relationships between colonization of S. aureus and deterioration of ICD. To this end, we first topically applied S. aureus to wild-type murine ears as a skin colonization model, which by itself did not induce clinical and histological inflammation. Nevertheless, flowcytometric analysis showed infiltration of neutrophils with increased CD49d expression in the S. aureus colonized skin. Next, we evaluated the susceptibility of S. aureus colonized skin to inflammation using an ICD model. S. aureus colonized skin showed stronger ICD responses compared to non-colonized skin. With use of confocal microscopy, more neutrophil extracellular traps (NETs) formation was detected in the S. aureus colonized skin than in the non-colonized skin after ICD induction. In addition, neutrophil depletion by anti-Ly6g antibody suppressed the exacerbated inflammation by S. aureus. Consistently, inhibition of NETs formation ameliorated the subsequent ICD exacerbation. These results suggest that S. aureus colonization itself does not induce clinical or histological inflammation, but induces infiltration of activated neutrophils, which subsequently promotes NETs release and exacerbates inflammation under irritant stimulation. Therefore, suppression of neutrophil accumulation and NETs formation may be a new therapeutic target for ICD such as hand eczema in AD patients." @default.
