• W4366245875 abstract "The present study was carried out to systematically evaluate the regulation of fillers on tablet properties of different natural plant product (NPP) powders. In our previous research, NPP powders were categorized into five groups according to their tabletability. The category II ∼ IV NPP powders and four fillers with different compression mechanisms were selected for research, including microcrystalline cellulose (MCC), lactose, starch, and dicalcium phosphate anhydrous (DCPA). The recommended formulation and design space for category II and category IV powders were obtained by mixture design. On this basis, the formulation of the category III powder was inferred and verified. The results showed that: (i) Category II powders had better tablet properties and larger design space compared to category IV powders, which further substantiated the reliability of the previously reported classification method. (ii) For category II powders, a combination of plastic and brittle deformation excipients was recommended, including the combination of 14.5% MCC, 3% lactose, and 9.5% DCPA or 12.9% MCC and 14.1% DCPA. (iii) For category IV powders, it was recommended to use plastic excipients with the addition of some brittle plastic excipients. The recommended filler was 27% MCC, or 13.5% MCC and 13.5% lactose. (iv) The recommended filler for category III powders was a combination of MCC and lactose in ratios of 1:1 – 4:1. And the verification results of the above formulations were satisfactory.
