- W43672175 abstract "Sepsis often results in severe pulmonary dysfunction. Via the thoracic duct, the lung is the first organ exposed to gut derived mediators released into mesenteric lymph during sepsis. Recently, it has been shown, that an enteral immunonutrition with olive oil significantly reduced the inflammatory mediator release from the gastrointestinal tract during sepsis and consecutively reduced septic pulmonary dysfunction. Aim: To investigate whether apolipoprotein (apo) A-IV, released from enterocytes during lipid absorption and known to have an anti-inflammatory potency, improves pulmonary dysfunction during sepsis. Methods: Mesenteric lymph was obtained from lymph fistula donor wildtype mice (apo A-IV +/+) and apo A-IV knockout mice (apo A-IV −/−) during sepsis (LPS 5mg/kg i. p.) with and without enteral immunonutrition with olive oil. Sepsis lymph was then reinfused into the jugular vein of separate recipient wildtype mice. Thereafter the lung tissue was analyzed for distance of oxygen diffusion, inflammatory response and cell apoptosis. Results: Sepsis lymph induced a significant inflammatory reaction and apoptosis in recipient mice, whereas sepsis lymph collected during an enteral immunonutrition with olive oil induced only a mild inflammation but no apoptosis in the lung of recipient mice. In contrast, sepsis lymph collected from apo A-IV knockout mice during enteral immunonutrition with olive oil failed to reduce the inflammatory response and failed to prevent apoptosis in the lung of the recipient mice. Conclusions: Mediators in sepsis lymph induce pulmonary dysfunction. The lung may be protected by an enteral immunonutrition containing long chain fatty acids in source of olive oil. Apolipoprotein A-IV is essentially involved in mediating the anti-inflammatory effect of an enteral immunonutrition with olive oil during sepsis." @default.
