- W4382722015 abstract "본 연구에서 갈색거저리 유충 추출물(TMLE)의 두 가지 추출온도 조건 중 5℃ 추출물이 수율과 단백질 함유량 측면에서 우수하였으며, TMLE (5℃)가 면역 기능 소재로서의 활용 가능한지에 대해 평가하기 위해 대식세포(RAW264.7 cell)에 처리하였으며, cell viability, 면역 활성화에 영향을 주는 인자(cytokine, 세포 표면 활성 인자) 및 신호전달 경로에 대해 살펴보았다. TMLE (5℃)를 RAW264.7 cell에 처리함으로써 cytokine (TNF-α, IL-6), 주조직적합성복합체(MHC class II), 공동자극인자(CD80, CD86)의 발현을 TMLE (5℃)의 처리 농도 의존적으로 증가시켰다. 또한 MAPKs 인산화와 NF-κB 전사 기전에 의해 대식세포 내 면역 반응을 유도하는 것을 확인하였다. 따라서 갈색거저리 유충 저온 추출물은 MAPKs와 NF-κB 신호 전달 체계를 통해 대식세포 내 면역 증진을 유도하기에 면역 활성을 유도하는 기능성 소재로 사용될 수 있을 것으로 보인다. 본 연구에서 갈색거저리 유충의 저온 추출물이 면역 증강 소재로서 활용 가능성을 보여주지만, 향후 추출물 내 면역 증진을 일으키는 유효성분을 정확히 파악하고 분리 정제 과정을 거쳐 효능을 갖는지 검증하는 연구가 필요하다. Insect-based foods, such as Tenebrio molitor larvae, have gained attention owing to their potential health benefits. This study compared the yield and protein content of T. molitor larvae extract (TMLE) obtained at 5 and 75℃. The extract obtained at 5℃ had superior quality and was adopted for further analysis. The immunomodulatory activity of TMLE (5℃) on macrophages, including macrophage viability (RAW264.7 cells), cytokine concentration, and cell-surfacemarker expression, was evaluated. The TMLE (5℃) induced an immune response in macrophages through the mitogenactivated protein kinase and nuclear factor-κB transcription pathways, suggesting that TMLE (5℃) is a potential immune enhancer. No studies have reported the evaluation of immune activity with refrigerated extracts, making the findings of this study an important contribution to the field. They will have important implications in the development of novel immuneenhancing therapeutics." @default.
