- W4385282317 abstract "Estimation of time since death (TSD) is one of the most challenging aspects of forensic medicine. There are several methods used to estimate the TSD like physicochemical, entomological, biochemical, metabolic, autolytic, and physical methods, which provide a wide range of TSD, as they are affected by different factors, and these methods are always questionable. The approach behind the present study is to calculate the accurate TSD by using the fold change expression (FCE) of autophagy genes HMGB1, Bcl2, and Bax. Seventeen cadaver heart tissues were analyzed within a time frame of 0, 6, and 12 hours from the TSD, at different time intervals at room temperature. FCE was determined and the mean cycle threshold (Ct) values obtained were assessed using RT-PCR. FCE of HMGB1 by ΔΔCt method at different time intervals showed that FCE was upregulated in the first 6–9 hours after death and then almost stable till 15–18 hours and then again showed upregulation up to 24 hours after death. The Bcl2 and Bax show no FCE in the heart tissue at any point of time till 24 hours after death. FCE of HMGB1 can be used to determine PMI and can be useful for PMI estimation. However, Bcl2 and Bax showed no expression till 24 hours after death. These genes may be a reliable marker for determining the time since death in medico-legal cases, making the investigation of such new methodologies unique in their precision and objectivity in measuring PMI." @default.
