- W4385374566 abstract "Although yellow fever virus was the first positive-strand RNA virus to be studied, the first virus shown to be transmitted to humans by a mosquito bite, and the first RNA virus for which an attenuated vaccine was developed, little was known about the molecular characteristics of members of the genus Flavivirus until the 1980s. My scientific career progressed in parallel with the field of flavivirus molecular biology. The advancement of flavivirus molecular biology was slower than that of the picornaviruses due to characteristics such as a longer genome RNA, the virion lipid envelope, the inability to translate the entire polyprotein in vitro, and the instability of many full-length flavivirus cDNAs in bacteria. The completion of flavivirus genome sequences in the mid-1980s opened many new directions for advancing flavivirus molecular biology. Unique molecular features of flaviviruses discovered since then include conserved cis-acting genome RNA structures, sfRNA, a 5′ IRES, viral protein 3D structures and cellular protein interaction partners, viral RNA replication vesicle architecture, and the viral protein shell on the outside of the virion membrane. It is anticipated that future research on the functional consequences of sequence variations in flavivirus RNA structures and proteins will provide new insights." @default.
