- W4386772849 abstract "This chapter examines the isolation and structure determination of proteins. The study of a protein begins with isolating it from a natural source containing the target protein as a component of a mixture. The methods for protein purification include ammonium sulphate precipitation; size-exclusion chromatography; ion-exchange chromatography; affinity chromatography; and specific expression, often in <italic>E. coli</italic>. Meanwhile, the major experimental methods for determining protein structures are X-ray crystallography, nuclear magnetic resonance (NMR) spectroscopy, and cryo-electron microscopy. Computational methods for protein structure prediction from amino-acid sequence have recently shown very great improvements. The chapter then looks at the Critical Assessment of Structure Prediction (CASP) programme, homology modelling, and <italic>a priori</italic> structure prediction. Ultimately, a truly general and reliable method of structure prediction will revolutionize research on proteins." @default.
