The development of treatment methods has been hampered by the poor knowledge of the molecular pathways driving severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection. By using the high RNA-sequencing-based transcriptomics and systems biology techniques, our aim was to examine the molecular regulatory mechanisms behind the COVID-19 disease. RNA-seq data of Peripheral Blood Mononuclear Cells (PBMCs) from a group of 17 COVID-19 patient’s sample and 17 healthy samples (GSE152418) were analysed in order to create a gene expression matrix. To identify co-expressed key clusters in healthy control, weighted gene co-expression network analysis was used. Module preservation and module-phenotypic connections techniques were utilised to select important modules for differential co-expression network analysis. Then, to find hub genes, PPI network was built subject to co-expressed hub genes. The functional enrichment analysis was performed to investigate the potential function of genes inside the key modules. A total of 10 hub genes were identified as biomarkers under the “Turquoise” module. The GO and KEGG functional enrichment analysis identified that genes were significantly associated with chromosome condensation, histone kinase activity and spindle. Identification of hub genes, may help us to develop molecules, encoded by hub genes, as therapeutic targets for the prevention of COVID-19 infectious disease." @default.
