- W4387026105 abstract "Background: Atopic dermatitis (AD), a chronic inflammatory skin disorder with an onset commonly occurring during childhood, can have a significant impact on the quality of life of patients and their families. The concept of “atopic march” refers to subsequent development of other allergic disorders in children with AD including food allergy, asthma, allergic rhinitis, and hay fever. A family history of allergic disease has been shown to be an important risk factor for AD development in children. In this study, we identified early biomarkers (before AD onset) on the skin surface of children associated with future development of AD. Skin surface biophysical measurements and biomarkers were collected non- invasively from children (aged 3-48 months) from the following groups: (A) healthy with no family history of allergy (n=29), (B) healthy with family history of allergic disease (n=101), and (C) diagnosed with AD (SCORAD 4-8, n=30). Eleven children from group B were diagnosed with AD within 12 months following the measurements. The children who developed AD showed increased concentrations of certain skin inflammatory markers and microbial dysbiosis before disease onset, compared to those who did not develop AD in the same group and compared to those in the control group A. In group C (AD at baseline), lesional skin was characterized by increased inflammatory cytokine concentrations, a relative abundance of Staphylococcus aureus, increased transepidermal water loss rates, and decreased skin surface hydration. Defining risk susceptibility before AD onset through non-invasive methods may identify children who could benefit from early preventive interventions." @default.
