- W4387054346 abstract "TNFα converting enzyme (TACE) cleaves membrane bound TNFα (mTNFα) to its soluble (sTNFα) and more bioactive form, has been linked to cardiovascular disease and insulin resistance. PURPOSE: to investigate the effect of aerobic exercise training and/or diet induced weight loss intervention on skeletal muscle TACE in overweight older adults. Based on previously published associational data, we hypothesized that exercise training and/or weight loss would decrease TACE expression in skeletal muscle of overweight, relatively unfit older adults. METHODS: Thirty-three overweight (BMI: 33 ± 0.6 mL/kg/min), older adults (age:63 ± 0.8y, VO2max: 16.3 mL/kg/min) were randomized into four groups: control (CON, N = 9), supervised aerobic exercise training (EX, N = 10), dietitian-guided, diet-induced weight loss (DIET, N = 8), and diet + aerobic exercise training (DEX, N = 6). The interventions lasted six months. TACE expression (via Western blot), body composition, and VO2max were measured pre and post intervention. Data were expressed as percent change from pre-intervention. Comparisons were made using ANOVA. RESULTS: Percent change in skeletal muscle TACE was significantly different (p < 0.05) between CON (+16.1 ± 13%) and EX (-39 ± 13%), but not DIET (-11 ± 18%) or DEX (-18 ± 18%). Percent change in body mass was significantly different (p < 0.05) between CON (+0.5 ± 1.2%) and DIET (-6.5 ± 1.8%) and DEX (-8.9 ± 2.4%), but not EX (-0.6 ± 0.9%). Percent change in VO2max was significantly different (p < 0.05) between CON (-7.4 ± 3.8%) and EX (+13.8 ± 10.9%) and DEX (+26.6 ± 8.9%), but not DIET (7.0 ± 7.3%). CONCLUSION: The exercise training and diet-induced weight loss interventions were successful in improving VO2max in EX and DEX, and decreasing body mass in DIET and DEX.
