- W4387054711 abstract "Previous research examining the benefits of localized cold application to exercising muscle indicates altered gene expression post-exercise. However, it is unclear if the local cold stimulus or the combination of exercise and cold can explain these alterations in the muscle growth response (i.e., proteolysis and myogenesis). PURPOSE: Determine the independent effects of local cold application to the vastus lateralis(VL) for 4 hours at rest on markers associated with the human muscle growth program. METHODS: Participants (n = 12, 27 ± 6 years, 179 ± 9 cm, 82.8 ± 13.0 kg, 18.4 ± 7.1 %BF) applied a thermal wrap to both legs containing either circulating fluid (10 °C, COLD) or no fluid (room temperature, RT). Muscle samples were collected and processed to quantify mRNA (RT-qPCR) and protein (Western Blot) associated with myogenesis and proteolysis. RESULTS: Temperatures in the COLD were lower than RT for skin (13.2 ± 1.0 °C vs. 34.8 ± 0.9 °C; p < 0.001) and muscle (20.5 ± 1.3 °C vs. 35.6 ± 0.8 °C, p < 0.001). All proteolytic-related mRNA was higher in the COLD (FBOX32, p = 0.049; FOXO3a, p < 0.001; TRIM63, p < 0.001). Myogenic-related mRNA were lower in COLD (MYO-G, p = 0.001; MYO-D, p < 0.001); except for MYF6 which was greater in COLD (p = 0.002) vs. RT. No other myogenic associated genes were different between the limbs (MSTN p = 0.643; MEF2a, p = 0.424; MYF5, p = 0.523; RPS3, p = 0.589; RPL-3 L, p = 0.688). The phosphorylation : total protein ratio for a negative regulator for muscle mass was lower in the COLD vs RT (4E-BP1Thr37/46, p = 0.043), but no differences in mTORser2448 or p70S6K1Thr389 were observed (p = 0.509, and p = 0.579). CONCLUSIONS: The combination of higher proteolytic and lower myogenic related gene expression suggests more breakdown and less growth after 4 h of local cold application to the VL. However, the observed lower 4E-BP1 phosphorylation with cold would remove the inhibitory effects for muscle growth. Additional chronic cold application work is needed to determine the applied effects of cold application on muscle mass development." @default.
