- W4387062164 abstract "Cancer cachexia is characterized by unexplained weight loss accompanied by a decline in muscle strength and physical function. While the benefits of exercise countermeasures (EC) on improving functional capacity have been well acknowledged, their potential to combat loss of body weight (BW) and physical function under cancer-induced muscle wasting has not been fully elucidated. PURPOSE: To determine the clinical efficacy of different types of EC on alterations in BW and physical function in ApcMin/+ mice, a model of intestinal neoplasia. METHODS: Forty ApcMin/+ mice (6-14 wks of age) were randomly assigned into sedentary control (SC-Apc n = 10), aerobic exercise (A-Apc n = 10), resistance exercise (R-Apc n = 10), and combination of aerobic and resistance exercise (AR-Apc n = 10) for an 8-wk intervention period. The A-Apc group performed treadmill running at 18 m/min for 55 min at a 5% grade. The R-Apc group performed 3 sets of 4 repetition ladder climbs with weight attached to the base of their tail. The AR-Apc group performed the combined aerobic and strength program. BW and functional capacity were measured at pre-and post-intervention. Functional capacity included grip strength to measure force generation of forelimbs, wire hang to evaluate fatigue resistance, and inclined plane test to assess sensorimotor coordination of mice to maintain body position for 5 sec. Statistical significance was set at P ≤ 0.05. RESULTS: After the 8wk intervention, BW was significantly decreased in the SC-Apc (-11 ± 9%) and was maintained in the EC. Grip strength (-13 ± 8%) and wire hang time (-45 ± 12%) were significantly decreased in the SC-Apc, while no detectable changes were observed in the EC. Sensorimotor function significantly declined in the SC-Apc (-8.4 ± 7.5%) and was maintained in the A-Apc and AR-Apc. Only the R-Apc exhibited a significant increase in sensorimotor function (+2.8 ± 1.5%). CONCLUSION: Our data indicate that EC appear to preserve physical function and BW in tumor-bearing mice, suggesting that EC as a potential therapeutic strategy to counteract a wasting syndrome and physical impairments in cancer patients.
