- W4387260295 abstract "Research on trophoblasts can enhance our understanding of human embryonic development. However, the transcriptional characteristics of trophoblast differentiation under disordered conditions are limited understood. This study sought to compare the distinct transcriptional profiles of normal and abnormal trophoblast differentiation using the single-cell RNA-sequencing technique. We used two twin pregnancies with a complete hydatidiform mole and a coexisting live fetus (CHMCF) to identify the distinct transcriptional profiles of normal and abnormal trophoblast differentiation. In this model, the CHM and the coexisting fetus had the same gestational age and experienced the same uterine environment, providing natural and optimal conditions for a comparative study. Unsupervised clustering analysis was employed to identify the major cell types, and gene expression and cell composition were compared. 10 pairs of surgical specimens of CHM tissues and age-matched aborted normal human embryos were obtained at 6–11 weeks of gestation and performed immunohistochemical verification of differential proteins. In total, we obtained 32,989 cells from two patients. Bioinformatics analysis identified three trophoblast cell subtypes, cytotrophoblast (CTBs), syncytiotrophoblasts (STBs), extravillous trophoblasts (EVTs) based on heterogeneous cell states and functions. These functions included the over proliferation supported by metabolic alteration and the upregulation of gluconeogenesis in CTBs subpopulations of CHM. In STBs, abnormally expressed hormone genes were identified, which were validated in 10-pairs specimens. EVTs exhibited a loss of the vascular-invasive subtype involved in extracellular matrix formation with negative regulation. This study provides a detailed molecular and cellular portrait with insights into the specific role of trophoblasts in placental development. The findings have wide implications for functionally assessing trophoblast-induced pregnancy abnormalities and could provide a method for their early detection." @default.
