- W4387396366 abstract "We performed experiments to determine the specific effects of Matrix Metallopeptidase 9 (MMP9) on the contractile response in human myometrial tissues. Uterine myometrial tissues were obtained with informed consent and Institutional Review Board approval from term patients undergoing elective C-section. Myometrial portions were dissected into strips, suspended in Krebs solution at 37 °C, and connected to force transducers. Tissue strips were maintained under 2-3 g basal tension for at least 30 m and stimulated with 60 mM KCl. Uterine strips were then treated with 10 nM oxytocin for 20 m, followed by the addition of purified activated-MMP9 to 1 mg/mL (∼10 nM) or vehicle control (saline) for 20 m. Changes in isometric tension were recorded. LabChart software was used to calculate area under the curve and to determine contraction peak force and time between contractions. Groups that met assumptions of normality were compared by Student’s t test (area under the curve). The Wilcoxon signed rank test was performed to compare datasets with non-normal distributions (peak height and time between contractions). We observed a 20% increase in the contractile response as measured by area under the curve over time in tissue treated with MMP9 compared to a 12% reduction in the vehicle treated tissue (p=0.0078). We observed a reduction in peak height in MMP9-treated samples (p<0.001); however, the contraction frequency nearly doubled in MMP9-treated tissues (p<0.0001). The median time between contractions decreased after MMP9 treatment (from 4.1 m baseline to 2.6 m after MMP9 addition). In comparison, the median time between contractions increased in vehicle-treated tissues (from 3.7 m baseline to 4.6 m after vehicle addition). Addition of MMP9 is sufficient to enhance oxytocin-induced contractions in human myometrial tissues. MMP9 concentrations are elevated in plasma, amniotic fluid, and myometrial tissues from preterm laboring women and we have previously shown that inhibition of MMP2/9 reduces contractile responses to oxytocin in human myometrial tissues and delays parturition in mice. Together these data suggest elevated MMP9 promotes myometrial contraction and labor." @default.
