- W4387396587 abstract "There is evidence that the male reproductive organs contain a diverse resident population of microbes termed the microbiome. The microbiome is thought to be present in and different between healthy and diseased states. The advent of next generation sequencing has allowed characterization of the semen microbiome. We hypothesized that the semen microbiome profile would differ in men with non-obstructive azoospermia (NOA) as compared to that of fertile controls (FC) because prior work has shown alterations in the semen microbiome composition of men with male infertility conditions including obstructive azoospermia. We enrolled men with NOA (azoospermia on semen analysis, small testes volume on physical exam, and elevated serum FSH >10 IU/mL) and men with proven fertility (fathered at least one child) undergoing vasectomy. We collected semen samples from all participants. Semen samples underwent quantitative PCR and 16sRNA sequencing for microbial identification. A total of 33 men were enrolled which included 14 men with NOA and 19 fertile men. Alpha diversity was similar between the groups suggesting similar diversity within samples, whereas Beta diversity was different. This suggested similar diversity within samples and differences in taxa diversity between samples. At the phylum level, men with NOA had lower amounts of Proteobacteria and Firmicutes while Actinobacteriota was overrepresented compared to fertile controls. At the genus level, Enterococcus was the most abundant in both groups, while 5 genera differed significantly between the groups including Escherichia/Shigella, Sneathia, and Raoutella. Our study showed significant differences in seminal microbiome composition between men with NOA and fertile men. These results may suggest a functional dysbiosis that may be associated with NOA. Further research into the characterization and clinical utility of the semen microbiome is necessary including investigation into the causality of microbiome changes and infertility." @default.
