• W4387496776 abstract "Adolescent depression is common, and associated with a number of downstream poorer social, psychological and behavioural outcomes, many of which occur in early adulthood and then persist across development. However, the majority of these findings are associational, and may not be causal. Recent advances in genomic research, and the use of longitudinal data to help improve precision and temporal sequences can allow for more causal inference methods to be tested. We aimed to triangulate evidence using classic regression analysis and one-sample mendelian randomisation (MR) in a longitudinal population study to examine the causal effect of adolescent depression episodes on later adulthood. We used prospective measures of self-reported depression, assessed across adolescence (four waves) and a variety of early adulthood outcomes in individuals from the Avon Longitudinal Study of Parents and Children (ALSPAC; max n=4,411). Early adulthood outcomes included life satisfaction, general health, daily smoking, financial problems and not in education, employment or training (NEET) status. We examined associations between adolescent depression and outcomes through Poisson regression (association analysis) and then again using one-sample MR with genetic variants as instruments taken from the most recent GWAS of depression from the PGC MDD working group. Adolescent depression episodes were associated with all of the poorer downstream adult outcomes in the Poisson regression analysis - as to be expected. However, there was also causal evidence from one-sample MR that a greater number of adolescent depression episodes were associated with later poorer life satisfaction (beta=0.23, 95%CI: 0.13, 0.32, p=3.4 × 10-6), poorer general health (beta=0.2, 95%CI: 0.09, 0.30, p=9.4 × 10-5), and greater financial problems across early adulthood (beta=0.38, 95%CI: 0.20, 0.56, p=3.5 × 10-5). Causal estimates from one-sample MR for adolescent depression on early adulthood daily smoking (beta=0.34, 95%CI: 0.09, 0.58, p=0.006) and NEET status (beta=0.19, 95%CI: 0.07, 0.31, p=0.001) were weaker, but still supported the association analysis. We have shown that adolescent depression is likely to play significant role in shaping later adulthood outcomes, using both classical regression analysis, and through more causal inference methods that leverage the power of genetics, such as one-sample mendelian randomisation. Together, these findings suggest that preventing and successfully treating adolescent depression could mitigate some of the downstream effects on social, psychological and behavioural outcomes. More research is needed to determine whether the timing or just onset of adolescent depression is more important, and to also explore how these effects may persist later into the life-course." @default.
