- W4387642786 abstract "Bone marrow (BM) derived stem cells and peripheral circulating stem cells were implanted to induce angiogenesis in No-Option Critical Limb Threatening Ischemia (CLTI). Considering the potentiality of this therapy there are conflicting results related to BM harvesting methods that may affect stem cell concentration and quality. 75 patients with No-Option CLTI were treated with BM implantation. In 58 patients, BM was harvested using Harvest Technologies BMAC System (Group HT) with standard aspiration needle followed by automated centrifugation process, to produce BM Aspirate Concentrate (BMAC); in 17 patients, BM was harvested by Marrow Cellution Aspiration System (MC System) (Group MC). CD34+ cells/mL, CD117+ cells/mL, CD133+ cells/mL, CD309+ cells/mL, HCT (%) and BM purity (%) were compared between the two BM preparations. the retrospective analysis of a subset group age adjusted, show that quality of BM obtained using MC System is better, in terms of purity, compared to classic harvesting method prior to centrifugation. Harvested BM prior to centrifugation is characterized by a higher % of CD133+ cells compared to BM post centrifugation. Conversely, the MC aspirate had a larger amount of Very Small Embryonic-Like Cells as indicated by the higher % of cells CD133+, CD34+ and CD45-. These differences translate with decreased occurrence of leg amputations in Group HT compared to Group MC, and increase in TcpO2 in patients treated with BM aspirated using MC. BM manipulation, such as centrifugation, affects quality and number of stem cells with detrimental consequences on clinical outcome, as reflected by different amputation rates between the two groups." @default.
