- W44676162 abstract "This chapter discusses the experimental factors involved in separation by centrifugal elutriation. The chapter describes a few methods for preparation of single-cell suspensions for elutriation. Some cell systems, such as peripheral blood, are already in single-cell suspensions and require no treatment. Whole blood can be separated by elutriation with the red cells being removed in the elutriation process. Alternatively, erythrocytes and granulocytes may be removed by centrifugation through layers of Ficoll–Hypaque, and the remaining mononuclear cells separated by elutriation or buffy coat may be used to reduce erythrocyte contamination. The choice of temperature of elutriation is dependent on the ability to keep cells viable and free from perturbations that would affect the course of the particular experiments. In an elutriation experiment described in the chapter, testis cells dissociated from the tissue by trypsinization were elutriated in the presence of 0.5% Ficoll instead of BSA. The use of BSA and fetal calf serum in the medium has been compared in only one instance. Excessive aggregation occurred during early attempts to separate mouse spleen cells in medium containing fetal calf serum. The use of BSA instead of fetal calf serum appeared to reduce this clumping." @default.
