- W44776710 abstract "This chapter explores the presynaptic adrenoceptors and regulation of release. The chapter describes an early study in which an increase in noradrenaline release observed in the presence of alpha-receptor blocking agents was obtained in the range of drug concentrations that elicit postsynaptic alpha-adrenoceptor blockade. This led to the hypothesis that postulates the presence of alpha-adrenoceptors in the outer surface of the noradrenergic nerve terminals. These presynaptic alpha-adrenoceptors are involved in the regulation of noradren adrenaline release through a negative feedback mechanism mediated by the neurotransmitter itself. Consequently, noradrenaline released by nerve stimulation once it reaches a threshold concentration in the synaptic gap activates presynaptic alpha-adrenoceptors triggering a negative feedback mechanism, which inhibits further release of the transmitter. The magnitude of the inhibition of transmitter release during exposure to alpha-adrenoceptor agonists is inversely related to the frequency of nerve stimulation. The frequency dependence of the inhibition of noradrenaline release elicited by alpha-adrenoceptor agonists differs from the effects of neuron blocking agents that reduce the output of noradrenaline at high frequencies of nerve stimulation. The presynaptic dopamine receptors are of pharmacological rather than physiological importance in peripheral noradrenergic neurotransmission because exposure to dopamine blocking agents does not increase the stimulation-evoked release of noradrenaline as it is the case with alpha-adrenoceptor blocking agents." @default.
