- W48391605 abstract "Researchers at Washington University School of Medicine in St. Louis have worked to isolate and characterize uterine progesterone receptors. Specific progesterone-binding proteins were demonstrated in cytosol fractions of human endometrium and rabbit uterus and in the nuclear fraction of estrogen-primed rabbit uterus. Physiological response of the target tissue to progesterone to form a binding complex having a sedimentation coefficient of 8s is elicited only under very restrictive conditions (e.g. estrogen-priming). A competitive protein binding assay for progesterone which makes use of binding proteins from uterine cytosol fractions promises a higher specificity than is available with current assays which use plasma proteins. The binding proteins from rabbit uterus were characterized with the aid of sucrose density-gradient analysis. Among the various compounds tested which included estradiol-17-beta estrone estriol cortisol and testosterone 5-alpha-pregnane-320-dione most successfully competed with progesterone for binding sites on cytosol proteins. Binding behavior proved the progesterone receptors of human endometrium and rabbit uterus are distinguishable from plasma binding proteins.
