- W50847462 abstract "Lipid vesicles, or liposomes, have recently been shown to be a useful carrier system for the delivery of a variety of compounds to cultured cells. Ca2+-induced fusion of small phosphatidylserine (PS) vesicles, followed by chelation of the Ca2+ by EDTA, produces large unilamellar vesicles (LUV) consisting of a lipid bilayer and a large interior aqueous space. As these vesicles form from the cochleate cylinder Ca2+-PS complexes, they entrap ~10% of the aqueous phase, encapsulating solutes such as macromolecules and macromolecular complexes. After encapsulation by LUV, the double-stranded polynucleotide rIn·rCn associates with cells at a higher level, is less sensitive to degradation by cell surface enzymes, and induces production of the antiviral activity of interferon more efficiently than rIn·rCn free in solution. Poliovirus encapsulated by LUV is rendered insensitive to neutralizing antiserum and is capable of infecting cells that lack the poliovirus surface receptor necessary for normal infection. Parallel experiments using purified poliovirus RNA in vesicles show that the RNA can infect cells efficiently and that it is not degraded by external ribonuclease. These results suggest that LUV are capable of efficiently encapsulating macromolecules and delivering them to cells in a biologically active form." @default.
