- W5094865 abstract "In practice, the classic methods alone have proved to be inadequate to settle the question of whether enzymes utilize nucleophilic catalysis to catalyze phosphoryl and nucleotidyl group transfer reactions. Stereochemical analysis of these reactions has given the most important mechanistic information on the question of whether nucleophilic catalysis and covalent phosphoryl-enzyme or nucleotidyl-enzyme intermediates are involved in the mechanisms. From a purely chemical catalytic standpoint, enzymes do not require nucleophilic covalent catalysis to mediate phospho-group transfer. The most common mechanism is the single displacement with direct transfer between substrates bound at adjacent sites. Nucleophilic catalysis is limited to those cases in which the bond energy to the phospho group must be maintained during some other process, such as the interchange of acceptor substrates in a ping-pong pathway. ATP dependent synthetases are more complex, and phosphoenzymes can have more complex roles in a few of those reactions." @default.
