Under supervision of DWI ANDREAS SANTOSA and ANI SURYANI. Petroleum is the source of energy for industrial, transportation and domestic activities. As an energy resource, it has many advantages but also can cause environmental pollution. Bioremediation is a technology based on microbial activities which can degrade environmental contaminants. Addition of surfactants along with bacterial inoculation have been reported to enhance the degradation process. The purpose of this research was to determine the effect of Bacillus sp. strain ICBB 7859 and ICBB 7865 and the addition of surfactant on the biodegradation of crude oil in liquid minimum medium and non-sterile oil contaminated soil. In 100 ml of liquid minimum medium the inoculants were inoculated at the population density of 1.00 x 10 cells/ml while in 200 g of unsterilized soil and crude oil mixtured (10% wt/wt) 1.00 x 10 cells/g. The dosage of surfactant Tween 80 was at its critical micelle concentration, i.e. 0.015 ml/l. Inoculation treatments with Bacillus sp. strain ICBB 7859 and ICBB 7865 in liquid minimum medium along with addition of surfactant enhanced crude oil biodegradation. Inoculation treatments with Bacillus sp. strain ICBB 7859 and ICBB 7865 on oil contaminated soil resulted lower total petroleum hydrocarbon (TPH) and pH, and higher biodegradation and CO2-C production than without bacteria inoculation. However, addition of surfactant treatment resulted lower CO2-C production than without addition of surfactant at 28 day of incubation.
