The beneficial effects of soil organic matter and humus on plants stem from the interaction between microbes and organic matter. Microbes are the mediators of carbon and nutrient turnover; microbes, together with abiotic reactions, mediate the formation of humus. Humus affects the survival and efficiency of the microorganisms. Even cellular metabolism during respiration, photosynthesis, and nitrogen fixation is affected by humus. Various techniques that determine the microbial biomass are discussed in the chapter. Traditional methods like direct observations under the microscope are tedious but with some recent modifications like special dyes, allow distinguishing between living and dead, or between active and inactive organisms. Direct biomass determination is carried out by the chloroform-fumigation-incubation method. Substrate-induced respiration and chloroform-fumigation-extraction methods have become the most widely adopted methods." @default.
