- W561185196 abstract "1383 Objectives Specific radiotracers for image osteoarthritis in cartilage are not established yet. Glucosamine sulfate (GS) is widely used in treatment of osteoarthritis, therefore could monitoring its uptake after radiolabeling. Methods We performed radiolabeling of GS using 99mTcO4-/tin method and investigated its uptake in degenerated cartilage. For uptake articular surgery tissue from individuals (n=4, 68-79a) undergoing knee arthroplasty (pieces of ~3mg) were used. The uptake of 99mTcGS (10µCi) was monitored from 0.5 to 72h to achieve the saturation. By some individuals was possible to get better preserved areas of tissue and to compare their 99mTcGS uptake with areas of heavy degeneration from the same patients. Degeneration of both areas was examined histologicaly using hematoxylin-eosin staining. 99mTcGS uptake was compared with histological status. 99mTcGS uptake prior to incubation with cold GS was investigated too. Results The optimal radiolabeling was obtained by using 140-160 MBq of 99mTc/0.1mg tin, pH 7. Specific activity amounted to 21.6-27 mCi/mM, n=4. The uptake of 99mTcGS was time-dependant increasing from 10 min. up to 72h , reaching the saturation at 24-72h. The maximal uptake in all investigated individuals was in range 22.50-74.6%, at saturation. The non-specific uptake in the presence of 50-fold excess of unlabeled GS was increasing with time up to a max. of about 15%, at saturation. We found a gut correlation of 99mTcGS uptake with cartilage degeneration by 25.0-32.4% increase in heavy, versus moderate degeneration. This was paralleled by histological examination. In better preserved tissue only cartilage surface was altered, while by heavy damage deep fisures and clefts were seen. Also by preincubation with cold GS, additionally 99mTcGS uptake in heavy damage was strongly reduced by 35.1-50.2%. Conclusions Because of high and specific uptake, 99mTcGS could be a suitable tracer to quantify degeneration in articular cartilage.
