- W57239145 abstract "A series of experiments were undertaken to examine the amount of protein recovered by an extraction medium from defatted soy flour. The effects of temperature, pH, agitation, sodium bisulphite, extraction ratio and time were subsequently studied. Defatted soy flour with a protein content of 53.5 ± 0.4% was the raw material for the experiments. The soy protein was extracted from the soy flour with an extraction medium prepared with water with added sodium hydroxide and sodium bisulphite. The resultant protein extract was analysed for protein concentration. Subsequently the spent flour was re-extracted with fresh extraction medium and analysed for additional protein extracted. All components in the extraction and re-extraction were measured to undertake material balances to check on data consistency and possible losses. The material balances showed that approximately 7 % of the initial protein was unaccounted for in the extraction process and for the re-extraction process. This is probably within experimental error. There was a discrepancy in the estimate of the inerts (fibre) in the material balances.Subsequent experiments were undertaken to attempt to extract more protein. The conditions altered in the subsequent experiments were to extract at 4oC and 50oC temperature, pH of 7 and 9.5, a larger extraction ratio of 16.7: 1 and lastly no agitation within a 24 hours extraction time. Also a study was made to compare the effects on the protein extract with and without sodium bisulphite added to the extraction medium.The subsequent experiments with the changes in the experiment conditions did not produce significant changes in the extraction results. In all cases approximately 60% of the protein was extracted, which is similar to Lui’s (2005) result. Therefore it can be confirmed that none of the conditions altered are a dominant variable in the extraction process.On the other hand, sodium bisulphite was found to be an important variable in the process. The initial extraction was higher at 32.6 mg/ml (~ 61%) with sodium bisulphite compared to 27.4 mg/m1 (~ 51%) without it. However there was a significant drop in protein concentration for extracts with and without bisulphite stored for several days." @default.
