- W577722319 abstract "We propose an extracellular optical recording configuration for neural signal recording by using Surface Plasmon Resonance (SPR). The SPR method is a nonlabeling technique, so that this makes great advantages for neuronal signal detection over conventional optical approach based on voltage-sensitive dyes: for example, a measurement time is not limited by fluorescence decay, and neurons are safe from potential toxicity of fluorescence dyes. Previously, we investigated optical performance of the SPR method to measure voltage-modulation effect at goldelectrolyte interface. However, such experiment did not exhibit good signal quality applicable to neuron action potential measurement. One of the major reasons was a noisy environment of optical recording configuration (laser beam fluctuation). In addition, biocompatibility of optical glass with high refractive index (SF-10, n=1.723 at 633 nm wavelength) was not guaranteed since primary cultured neurons were not viable onto gold-coated SF-10 glass slides. Here, we introduce an improved optical configuration with better signal-to-noise ratio by replacing 1) light source with a low noise laser diode and 2) electronics (filter and amplifier). The improved performance was confirmed by current stimulation into an aqueous medium over gold-coated SPR chips by using an external current source. This was done under similar consideration with conventional microelectrode performance test for neural signal detection. We modified the SPR system to use a low refractive index glass (BK7, n=1.515 at 633 nm wavelength). Primary cultured neurons were viable for more than 2 weeks on goldcoated BK7 glass slides. The experimental consideration for in-situ SPR recording with cultured neurons will be discussed." @default.
