- W58245112 abstract "Idiosyncratic drug-induced liver injury (DILI) is a major challenge in pharmacotherapy and drug development. The precise mechanism underlying this rare drug toxicity is unknown, but is believed to involve a variety of environmental and host genetic factors. Genetic variation in drug metabolism and drug transport has been suggested as a contributing factor. Recent genome-wide association studies of DILI have revealed a major role for human leukocyte antigen (HLA) genes in DILI risk, suggesting that adaptive immune responses play a key role in the development of DILI for some drugs. While there are currently no widely accepted DILI-predictive biomarkers, future studies may provide clinically useful genetic and nongenetic markers by taking advantage of the continuing improvements in genomic and epigenomic technology." @default.
