• W591515279 abstract "Peripheral blood lymphocytes (PBLs) from HIV seropositive patients, cultured in growth factor free medium for 3 days show an important loss of viability and an apoptosis phenomenon in T cell subsets compared to healthy donor’s PBLs. Different reports describe a decrease of intracellular Glutathione (GSH) in T lymphocyte subsets correlated to the stage of AIDS disease. N-Acetyl-cysteine (NAC) has been described to increase Glutathione level and restore cellular redox pathways. 15 patients were treated with 600 to 1200 mg of NAC per day over a year. Viability and apoptosis profiles were followed over a 10 day period. In a flow cytometry (FCM) test using Acridine Orange, we noted a cell population with low DNA staining intensity in PBLs from HIV seropositive patients. Agarose gel electrophoresis revealed DNA fragmentation correlated to this phenomenon, suggesting apoptosis. With NAC treatment we observed a systematic and stable amelioration of viability results from 2 months to 6 months of NAC treatment. At day 3 no cell death or apoptosis is detected in PBLs from healthy donors and patients treated over 6 months. At day 6 PBLs from healthy donors have up to 30% dead and apoptotic cells (60 to 100% for non or recently treated patients). There is no cell death before day 8 to 10 in PBLs from patients treated over 6 months. N-Acetyl-Cystein administration to HIV seropositive patients seems to prevent early cell death and to lead an “overprotection” of patients’ PBLs." @default.
