- W596647164 abstract "Dietary Flaxseed (FS), a nutritional supplement with antioxidant and anti-inflammatory properties, abrogates radiation-induced pulmonary fibrosis in a mouse model of radiation pneumonopathy. In this study we evaluated the genetic profile of dietary FS in lung to detect changes in gene expression that might elucidate the protective action of FS. Mice (n=6–8 per group) were fed a 0% or 10% FS diet for 3 weeks followed by a single fraction thoracic XRT (Groups R-0% and R-10%). Mouse lung mRNA was isolated 48 hours post XRT and microarray analysis performed on 30,000 genes. Non-irradiated mice were also evaluated for each diet (Groups C-0% and C-10%). Three datasets were generated from comparisons among groups with p<0.05 and Absolute Ratio>1.2. Dataset A (effects of FS in XRT) identified 1218 genes differentially expressed between R-0% and R-10%: 45% were up- and 55% down-regulated. Dataset B (effects of FS alone) identified 1512 significant genes between groups C-0% and C-10%: 56% were up- and 44% down-regulated. Dataset C (effects of XRT alone) identified 3278 genes between groups C-0% and R-0%: 56% were up- and 44% down-regulated. Three-hundred four significant genes were identified from (A minus B) intersect C, representing only genes modified by both XRT and FS. Remarkably, 93% of these genes had reversed their expression back to baseline as a result of the FS diet, providing useful insight into the possible mechanisms of FS action in lung injury.
