Osteoblasts play an important role in the formation of the calcified matrix and appears to be the target cells of many hormones, such as parathormone (PTH), calcitonin, and 1,25-dihydroxycholecalciferol. The purification of osteoblasts helps elucidate the mechanisms of bone formation, remodeling, resorption, and metabolism. The development of methods of bone cell isolation has greatly advanced the study of metabolism of bone tissue. Enzymatic digestion is one of the most advanced techniques for isolating bone cell populations and obtaining osteoblasts. Although enzymatic isolation procedures have helped the studies on the biological and biochemical mechanisms of osteoblasts, these mechanisms can be identified more precisely only with osteoblasts free of any contamination of other bone cells. By using the cloning method, some kinds of heterogeneous components of bone tissue can be purified by several culture passages, and the pure component will retain its morphological and biochemical characteristics." @default.
