- W600070106 abstract "Background: Attention deficit hyperactivity disorder (ADHD) is a common neuropsychiatric disorder in children. This disorder is characterized by significant behaviors like inattention, hyperactivity and impulsivity. Psychostimulants as methylphenidate (MPH) are the most effective and widely used treatment for ADHD. Little is actually known about the action of MPH but it is believed that it works by blocking the reuptake of noradrenaline and dopamine and by stimulating the release of these catecholamines. The aim of the present study was to investigate the effect of chronic MPH on dopamine D1 receptor protein in different parts of the brain in young rats.Method: For the measurements of D1 receptor protein levels, western blotting followed by optical density measurements using an image analysis system was used. Protein values were determined in different brain regions at 2 and 24 hours after the last administration. Young male rats (20 days old) were treated with MPH 2 mg/kg (i.p.) or saline 1ml/kg, twice daily for 2 weeks.Results: Compared to the corresponding saline treated control groups, treatment with MPH showed no significant effect at 2 or 24 hours after the last injection in any of the brain regions analysed. However in nucleus accumbens MPH did show a noticeable non-significant decrease of D1 receptor protein levels at 24 hours after the last injection.Conclusion: The present study demonstrated that there was no significant difference of D1 protein levels in any of the brain regions measured. Interestingly, a previous study has also shown a non-significant decrease by MPH on D1 receptor expression in the nucleus accumbens." @default.
