- W604819077 abstract "The transition metals, copper and iron, are well known to catalyze the generation of free radicals in vitro and may be involved in the pathogenesis of various diseases. Wilson’s disease is an inherited disorder of copper metabolism characterized by progressive accumulation of copper in the liver and its subsequent overflow to extrahepatic tissues. Non-ceruloplasmin copper is increased and ferroxidase activity is decreased in the plasma. The copper released from the liver into the circulation catalyzes the generation of free radicals, which consume radical trapping antioxidants in the plasma, such as urate, vitamin C and vitamin E. The radical trapping capacity in plasma of Wilson’s disease is decreased at the early stage of the disease without therapy, and it is improved by copper chelating therapy with d-penicillamine and tirientine. In LEC rats (a model of Wilson’s disease), the plasma antioxidant activity against ferrous salt-stimulated lipid peroxidation is reduced, and addition of zinc inhibited the peroxidation. This finding provides a basis for the improvement of Wilson’s disease by zinc therapy. Homocystinuria is an inborn error of methionine metabolism characterized by elevated levels of homocysteine in the body fluids which cause premature atherosclerosis and thrombosis. Transition metal-chelated oxidation of homocysteine thiols is thought to lead to the generation of superoxide and hydrogen peroxide. Active oxygen species generated in plasma appear to attack lipoproteins to produce oxidatively modified LDL, which may contribute to the onset of atherosclerosis.
