- W6069414 abstract "Objective:To investigate collagen type Ⅰ and type Ⅲ and their mRNA expression in rat models of abdominal aorta coarctation.Methods:14 of 30 SD rats were taken out randomly as sham operation group,and another 16 rats were selected as model group.Rat models were established by partially banding abdominal aorta with silver clip(0.7 mm of inside diameter) according to methods used by Doering's.Eight weeks later,collagen type Ⅰ and type Ⅲ were stained by immunohistochemistry and the optical density values were measured subsequently as a marker of their content.The mRNA expression levels of collagen type Ⅰ and type Ⅲ were analyzed after real-time PCR amplification.Results:In the 8th week,average optical density values and the mRNA expression levels of collagen type Ⅰ and type Ⅲ in model group increased significantly compared with sham operation group(P0.01).Immunohistochemistry stain displayed that collagen typeⅠ and type Ⅲ in myocardium stroma of model group increased,thickened and disorganized remarkably.Conclusion:The mRNA expressions of collagen type Ⅰ and type Ⅲ in fibrotic myocardium induced by abdominal aorta coarctatio appear to increase mainly in intercellular substance and perivascular space.This model is suitable to study myocardial fibrosis during chronic process of some diseases,such as heart failure,hypertension and so on." @default.
